Pick two molecules in your biological system that bind to one another
Build a model for this Gedanken experiment:
Cuvette, 10000 molecules of one, 100000 molecules of the other added at t = 10 sec. Estimate the Kd. Set k1 and k-1 accordingly.
Plot both free concentrations and the complex concentration
After equilibrium, add an antibody or chelator that removes all of the free added molecule
How much of the added molecule is free at the final equilibrium? Go home when you’re done (grin).
